The company was formed 1962 with a DC-3 and some light aircraft. In 1964 Curtiss C-46s were added to the 2 DC-3s and cargo flights to San Juan and Miami were operated.
In December 1966 a L-749A was purchased and in 1967 a L-049 was leased. In the coming years Qusiqueyana built up a daily passenger service between Santo Domingo and Puerto Rico. In 1975 2 L-049s were added and put into service. Transatlantic charters with Boeing 707s were not successful and the services ended in 1976. With the withdrawal of the passenger transportation permit to fly passenger in Constellations in January 1978 the company flew the world’s last passenger schedules with a L-749A to and with a L-049 from San Juan to Santo Domingo. Though flying cargo services Quisqueyana ceased operations in April 1978.
